A classical experimental framework comprising criteria intended to link a specific microorganism to a specific infectious disease: the agent should be present in every case of the disease, isolatable and grown in pure culture, cause the disease when introduced into a susceptible host, and be recoverable from that experimentally infected host. These criteria describe an idealized proof model and do not apply universally.

Applying the postulates rigidly to agents that cannot be cultivated in artificial media, to diseases with asymptomatic carriers, to obligate intracellular pathogens, or to human‑restricted infections where ethical inoculation is impossible; doing so can lead to false negatives or rejection of valid causal evidence.

When applicable, they guided experimental demonstration of pathogenicity and remain a conceptual touchstone for pathogen identification; misconstrual as universally applicable discourages use of molecular, epidemiologic, or ethical evidence needed for many modern pathogens.
